Area B: Biological Targets B2: Probing mechanisms of the Cdc48/p97 segregase with designed supramolecular ligands
B2 will probe three prominent structural elements of the segregrase p97 with customized supramolecular compounds to explore their biological function: the central pore, adapter binding and the N-D1 communication will be examined in biochemical and cell-based assays to understand their relevance in p97 function. In addition, supramolecular p97 ligands and p97 itself will be delivered into mammalian cells using CaP nanoparticles to study uptake and targeting of intracellular p97 in different subcellular localizations.
