Die Herpes Simplex Viren vom Typ 1 (HSV-1) und Typ 2 (HSV-2) gehören zu den häufigsten Krankheitserregern beim Menschen. Bei Patienten mit intaktem Immunsystem können sowohl Primär- als auch chronische HSV-Infektionen schmerzhafte Läsionen im Oral- bzw. Genitalbereich verursachen. Diese sind jedoch lokal begrenzt und lassen sich in der Regel gut mit den verfügbaren Medikamenten behandeln.
Schwerwiegende und zum Teil lebensbedrohliche Komplikationen sind bei Infektionen im Augenbereich sowie bei Patienten mit Immunsuppression zu erwarten. Häufig wiederkehrende HSV-Infektionen am Auge können beispielsweise zur HSV-1 Keratitis führen, die zu den führenden Ursachen von Erblindung in der industrialisierten Welt zählt. Bei Patienten mit Immunsuppression können HSV-Reaktivierungen zu generalisierten Infektionen führen, die ohne wirksame Behandlung tödlich sind. Des Weiteren führt die Übertragung des Virus auf Neugeborene zu lebensbedrohlichen Infektionen. Die Ausbildung von resistenten Virusstämmen erschwert die Behandlung zunehmend. Neuartige Ansätze zur Prävention und Therapie von HSV-Infektionen sind daher dringend erforderlich.
Im Rahmen unserer Forschungsarbeiten beschäftigen wir uns daher mit der Entwicklung neuartiger Strategien zur Vorbeugung und Behandlung von HSV-Infektionen. Wir konnten einen humanisierten Antikörper entwickeln, der bereits erfolgreich bei immunsupprimierten Versuchstieren sowie im Mausmodel der Herpes-Keratitis getestet werden konnte. Gegenwärtig arbeiten wir an einem Nanopartikel-basierenden Impfstoff, der die Induktion von neutralisierenden Antikörpern gegen HSV vermitteln soll. Diese Antikörper sollen zudem in der Lage sein, die direkte Virusausbreitung über den „cell-to-cell spread“ zu inhibieren. Diesen Mechanismus benutzen Herpesviren, um der Immunantwort des Menschen zu umgehen. Das Ziel unserer Arbeit ist es, neue Erkenntnisse über die Prävention von HSV-Infektionen zu gewinnen, die zur Entwicklung eines erfolgreichen Impfstoffes beitragen sollen.
